State-of-the-art equipment and sophisticated analytical chemistry techniques are provided by the Analytical Chemistry Shared Resource to cover a broad variety of needs. Personnel are trained experts in the operation and maintenance of this equipment, as well as having the expertise to develop and optimize the appropriate methods. Available equipment includes:

Liquid Chromatography-Mass Spectrometry (LC-MS)

LC-MS instrumentation includes a ThermoElectron TSQ Quantum Ultra and a TSQ Quantum triple-quadrupole mass spectrometer with electrospray and atmospheric pressure chemical ionization. These instruments are the centerpiece of the laboratory and represent some of the most technically advanced triple-quadrupole technology available today. They are used for both quantitative and qualitative analysis of small molecules in biological samples. The sensitivity and selectivity of their design makes it ideal for measuring drug and biochemical marker levels in complex biological matrices. The linear dynamic range of the triple-quadrupole-based mass analyzer makes this the ideal instrument for quantitative analysis, especially for those studies involving pharmacokinetics.

Gas Chromatography-Mass Spectrometry (GC-MS)

GC-MS instrumentation includes a ThermoElectron Trace DSQ, containing a computer-controlled GC coupled to a single-quadrupole mass spec detector with electron ionization (EI) and chemical ionization (CI) available in either positive or negative ion modes. This instrument is used mainly for quantitative analysis of volatile compounds such as eicosanoids and lipids. The extreme sensitivity of GC-MS makes it the technique of choice for volatile and very low mass compounds.

Atomic Absorption Spectrophotometry (AAS)

AAS instrumentation includes a Perkin-Elmer Aanalyst 600 AAS with graphite furnace and Zeeman background correction. This instrument is used exclusively for analysis of selenium, gold and platinum metals, although it has the capacity for analysis of many other metals. The ability to measure selenium levels in biological samples is an important feature of the Analytical Chemistry Shared Resource.

High Performance Liquid Chromatography systems (HPLC)

HPLC instrumentation consists of Waters 600 series equipment with UV-Vis diode array, UV, and fluorescence detectors. These are workhorse systems that are used extensively when fluorescence detection is required or UV detection will suffice and mass spectrometric detection is not needed.

Ultra Performance Liquid Chromatography – Quadrupole/Time of Flight Mass Spectrometry (UPLC-QTOF)

The Waters UPLC-QTOF was recently acquired for metabolomics analyses of clinical samples.  Thousands of metabolites can be detected in a single sample.  The instrument features separation using the Acquity UPLC I-Class System which achieves 8x faster separation times and 2.5x the sensitivity over HPLC at the same resolution.  The UPLC is coupled to the Xevo G2-S QTOF which has the basic footprint of a quadrupole/time of flight mass spectrometer.  The StepWave ion optics filters neutral species and allows charged ions from the source to be focused to the MS analyzer, increasing sensitivity.  A single quadrupole allows for MSE data acquisition and MS/MS experiments.  Up to 30 spectra/sec can be detected while maintaining a resolution of >32,000 FWHM (full-width at half mass). The TransOmics Metabolomics software features “Chemically intelligent” tools for data reduction, interpretation, metabolite identification, and integration of identified metabolites into biological pathways.
